Tutorial: Using the Basic Tools in Photoshop
When you open Photoshop, you see the following screen:
1. If you want to place the new file on your computer’s desktop, choose Edit⇒Open or simply press Ctrl+O.
2. If you want to open the file right away, you can double-click its icon. Either way, you see the Photoshop interface.
3. Drag the file’s icon to your desktop. Then switch to the window on the right side of your desktop and open it by double-clicking its icon. Then choose Open.
4. Create a new image by choosing File⇒New or pressing Ctrl+N.
5. Choose File⇒Save As.
6. In the Save As dialog box that appears (see Figure 2-1), give the new file a new name, and save it in the folder that has been created for your Photoshop files on your computer.
To save a file under any name you want, you don’t need to use the Save As command. You can simply right-click and choose Save.
7. Choose File⇒Close.

8. In the Graphics window, you see a new image file displayed.
9. Open it by double-clicking its icon.
10. Create a new layer by clicking the New Layer button in the top-left corner of the Photoshop window, just below the New Window button.
For new layers, Photoshop displays the name _Untitled 1_ in place of the default name, which appears only on layers that contain more than one image.
11. Click the New Layer button again to create another layer.
12. Fill the new layer with black by choosing the Fill or Color box and clicking the Fill tab and the color box, as shown in Figure 2-2.
13. Create another layer by clicking the New Layer button.
14. Fill this layer with the color you want to change. Figure 2-3 gives you some examples.
15. Choose the Edit tab and then click the Eraser tool to remove the black fill.

Inhibition of human and rat liver ornithine decarboxylase by alpha-methylarginine.
The specific interaction of alpha-methylarginine with human and rat liver ornithine decarboxylase (ODC) was studied using purified human and rat ODC, respectively. In the rat, two complexes between the ODC and its inhibitor were observed. One complex (M2) is formed with the reaction of the enzyme with both alpha-methylarginine and with the inhibitor alpha-methylornithine. The second complex (M1) is formed by reaction of the inhibitor alpha-methylornithine with purified human ODC. Alpha-Methylornithine inhibited purified human ODC with the formation of a very tight complex, similar to that formed with the rat enzyme. This complex, however, was not isolated by gel filtration chromatography. An analysis of the interaction of alpha-methylornithine with human and rat ODC by gradient gel filtration has shown that alpha-methylornithine promotes a decrease in the association constant between ODC and the enzyme. In contrast to the inhibition of human and rat liver ODC by alpha-methylornithine, alpha-methylarginine showed a dose dependent inhibition of purified human ODC. A purification procedure for alpha-methylarginine was designed in order to obtain this compound in high yield. Two different molecular species were isolated, one of which was alpha-methylarginine itself. When the inhibitory properties of alpha-methylarginine were compared with those of alpha-methylornithine, in the case of purified human liver ODC it was found that alpha-methylarginine is a more potent inhibitor of the enzyme (Ki = 19.2 mM vs 11.7 mM for alpha-methylornithine). In contrast, in the case of purified rat liver ODC, alpha-methylarginine was less potent than alpha-methylornithine (Ki = 34.2 mM vs 24.4 mM). Since no significant complex could be observed between purified human liver ODC and alpha-methylarginine, it may be concluded that this inhibitory interaction is not an absolute requirement for the complex formation between rat liver ODC and alpha-methylornithine.Lloyd Smith (Australian politician)
